2,3:5,6-Di-O-isopropylidene-α-D-mannofuranose is a furanose sugar derivative. It contains α-D-mannofuranose whose hydroxyl groups at positions 2, 3, 5 and 6 are protected as isopropylidene acetals. This renders the furanose ring rigid and prevents its interconversion to other hemiacetal forms. It is a colourless crystalline solid. 2,3:5,6-Di-O-isopropylidene-α-D-mannofuranose is widely used as a protected building block in organic synthesis of complex carbohydrates and natural products. The acetal groups block reactivity at the hydroxyl positions, allowing selective monofunctional transformations like esterification and etherification to be carried out on the remaining hydroxyl group. This helps prevent side reactions and simplify product isolation. It is employed to synthesize various mannose-containing glycosides, glycoconjugates and oligosaccharides found in nature. It also serves as a starting material to prepare nucleosides, antibiotics and antitumor agents through glucosylation, amination and oxidation reactions. As the furanose ring is locked by the acetals, it is a useful substrate for studying carbohydrate transformations and anomeric effects. This reagent further aids in developing novel carbohydrate-based polymers with applications in biomedicine and bioengineering.
