Alkaline phosphatase-conjugated secondary antibodies are used to bind to primary antibodies, and BCIP treatment generates a blue color in the presence of the enzyme, indicating protein localization within the tissue sample.
In situ hybridization: BCIP is also used for detecting nucleic acid sequences in fixed tissues or cells. Hybridization probes are labeled with alkaline phosphatase, and upon addition of BCIP, the presence of the target mRNA or DNA is indicated by the development of a blue color.
